Tests for cytotoxicity – respectively biocompatibility – are performed e. g. for biological evaluation of medical devices or during the development of microphysiometric systems. In the presented work a protocol for a chemically defined test for cytotoxicity is described. The chemically defined medium contained Waymouth’s Medium, 50 % DMEM, 50 % Ham’s nutrient, a mixture F-12 and one Insulin/Transferrin/Selen (ITS)-supplement (DME/F12 + ITS).

Sowohl für die Zellkultivierung als auch für den eigentlichen Test wird Natriumhydrogencarbonat-gepuffertes DME/F12 + ITS (0,5 μg/ml Insulin, 0,275 μg/ml Transferrin, 0,25 μg/ml Selen) mit 50 μg/ml Gentamicin (Sigma-Aldrich Chemie GmbH) verwendet. Das Medium wird auf pH 7,4 ± 10 % und eine Osmolarität von 300 mosmol/kg ± 10 % eingestellt und dann – vor Zugabe des ITS – sterilfiltriert.
