Scandinavian journal of immunology 65.6 (2007): 581-588. In this study, CB-CD34(+) (1 x 10(4)) cells were cultured for 8 weeks in a serum-free medium containing rhIL-6 and rhSCF with or without a human bone marrow stromal cell line, namely, co-culture and liquid culture, and were compared in various regards. Mast cells (MC) were basically determined by metachromatic staining of granules. The number of MC obtained, percentage of FcepsilonRI(+) cells, histamine content, and IgE-mediated histamine release were higher in the co-culture than in the liquid culture. The cells were cultured in serum-free QBSF-60.
