In this study, chemically defined serum-free culture conditions for the differentiation of embryonic stem (ES) cells into the PS-like cells was developed. Cultures supplemented with Wnt showed induction of expression of a PS marker, the brachyury gene, followed by induction of the anterior PS markers goosecoid and foxa2, a posterior PS marker, evx1, and the endoderm marker sox17. Similar differentiation of PS by Wnt was also observed in human ES cells. To culture mouse ES cells, ESF basal medium was supplemented with different components as specified in the Materials and Methods. The formulation of ESF basal medium is listed in a previous article, Furue et al. 2005. Human ES cells were cultured in hESF-dif medium. The chemical components of hESF-dif are same as those of mouse ESF except HEPES. HEPES is excluded from hESF-dif medium. Furue M, et al.
